چکیده

Previous studies had shown that the exogenous cannabinoids can induce anticonvulsion, which was believed to be mediated through the activation of central cannabinoid Type 1 receptors. Moreover, the endogenous cannabinoid anandamide has shown anticonvulsant properties in the in vitro preparations. The current study used 27 adult Sprague-Dawley rats to investigate the effects of another endogenously occurring cannabinoid called 2-arachidonyl glycerol on epileptiform activity induced by picrotoxin. Extracellular recordings were made from the pyramidal cell layer of the CA1 region of hippocampal slices maintained in an interface type recording chamber. Stimulation was performed by using single pulses. It evoked population spikes of approximately equal amplitude. Using single pulse stimulation, perfusion of 500 nM picrotoxin caused an increase in the amplitude of the first population spike, and caused epilepsy by introducing a second or multiple population spikes. In the presence of picrotoxin, 2-AG reduced the amplitude of the population spikes, thus reducing the epilepsy. The CB1 receptor antagonist, AM281 (500 nM) had no effect on responses recorded in the presence of picrotoxin, but totally blocked the effect of subsequently perfused 2-AG. The current results showed that 2-AG caused anticonvulsion in the in vitro preparations. The results will open avenues for cannabinoid receptors as possible future therapeutic targets.

Effects of arachidonyl-2’-chloroethylamide (ACEA) on the protective action of various antiepileptic drugs in the 6-Hz corneal stimulation model in mice

Accumulating evidence indicates that cannabinoid CB1 receptor ligands play a pivotal role in seizures, not only in preclinical studies on animals, but also in clinical settings. This study was aimed at characterizing the influence of arachidonyl-2'-chloroethylamide (ACEA-a selective cannabinoid CB1 receptor agonist) co-administered with phenylmethylsulfonyl fluoride (PMSF) on the anticonvulsant...

The role of BK channels in antiseizure action of the CB1 receptor agonist ACEA in maximal electroshock and pentylenetetrazole models of seizure in mice

The anticonvulsant effect of cannabinoid compound has been shown in various models of seizure. On the other hand, there are controversial findings about the role of large conductance calcium-activated potassium (BK) channels in the pathogenesis of epilepsy. In this study, the effect of arachidonyl-2′-chloroethylamide (ACEA), a CB1 receptor agonist, and a BK channel antagonist, paxilline, either...

Generation of diacylglycerol molecular species through the cell cycle: a role for 1-stearoyl, 2-arachidonyl glycerol in the activation of nuclear protein kinase C-betaII at G2/M.

Protein kinase C (PKC) is a family of 11 isoenzymes that are differentially involved in the regulation of cell proliferation. PKC-betaII, a mitotic lamin kinase, has been shown previously to translocate to the nucleus at G(2)/M and this was coupled to the generation of nuclear diacylglycerol. However, it is not clear how isoenzyme selective translocation and nuclear targeting is achieved during...

In-vivo Anticonvulsant and In-vitro Antitubercular Activity of methyl Indole Derivative of some 6-aryl-4, 5-Dihydropyridazin-3(2H)-ones and their Expected Anticonvulsant Mechanisms

Methyl indole derivative of some 6-aryl-4,5-dihydropyridazin-3(2H)-ones (3a-e) were synthesized by Mannich reaction and evaluated as anticonvulsant against MES (50mA, for 2sec), INH (250mg/kg), scPTZ (80mg/kg) and STR (3mg/kg) induced convulsion methods at 50 mg/kg dose level. All compounds 3a-e were also evaluated as antitubercular agent against M. tuberculosis H37Rv by MABA method. All compou...
